The GlobalFiler™ amplification kit is a multiplex test that identifies a set of 24 markers (STRs), allowing for the identification of human cell lines from genomic DNA. STRs are short repeated DNA sequences. The number of repetitions varies greatly among individuals and alleles. This technique should be used exclusively for research projects.

Technique

The identification is performed using the Globalfiler PCR amplification kit from genomic DNA.
The PCR kit includes specific primers for each marker. The resulting PCR will then be migrated by capillary electrophoresis on the DNA Analyzer 3730 XL (Applied Biosystems™). We use the analysis modules described in the Globalfiler user kit.

The kit includes an allelic scale control allowing for the automatic genotyping of most alleles for the kit’s loci and a positive DNA control for experiment validation.

The raw data is analyzed using GeneMapper.

    • Allows for easy evaluation of samples/lines among themselves by obtaining a percentage of homology.
