Titre de la conférence : Setting the pace: how barriers orchestrate neuronal maturation

Résumé : Neuronal differentiation is a highly orchestrated process where pluripotent stem cells transition into neural progenitors and ultimately mature neurons. Our research unveiled the temporal dynamics of gene expression, transcript usage, and splicing events underlying each stage of human neuronal differentiation. Notably, we identified a burst of transcriptional activity in a subset of genes during the progenitor stage, essential for successful neuronal differentiation. Disruption of these “bursting” genes impairs this process. Furthermore, progenitor-specific alternative splicing events were found to be critical for the maturation trajectory, acting as precise molecular switches that enable terminal differentiation. We uncovered a “barrier” in progenitor cells that holds transcriptional programs in a poised state, requiring release for neurons to acquire mature properties upon differentiation. Strikingly, this barrier is disrupted in neurodevelopmental disorders. In Fragile X syndrome, we demonstrated that the absence of FMRP interferes with key progenitor-stage molecular events, impeding the progression to mature neurons.
